I am currentlly trying to get quantitative results out of my western blot films. I used image J and Quantity One. However, though trends appear prominant to the eye, the results for quantification indicate insignificant difference. Variables like exposure to primary and secondary antibodies, development time, exposure to chemoluminescence, and washing are corrected but variability is still present. What are other variables that should be considered during quantification? Are there specific image details that should be considered? What are effective approaches to eliminate variability in image quantification?.
